Family with sequence similarity 53 member C (FAM53C) is a highly conserved, 392-amino-acid, low-complexity protein characterized by a flexible, intrinsically disordered structure with coil regions and short helices. While initially of unknown function, FAM53C was recently shown to bind directly to the protein kinase DYRK1A, anchoring DYRK1A in the cytoplasm and suppressing its kinase activity. This interaction modulates gene expression relevant to neurodevelopment and disease states such as Down syndrome. FAM53C may also indirectly impact cell cycle, cell proliferation, and transcriptional networks relevant to obesity and inflammation. FAM53C is not an enzyme, receptor, or transporter, and currently has no known drugs or compounds that interact with it directly
