Family with sequence similarity 72 member D (FAM72D) is a human protein belonging to the FAM72 gene family, consisting of four highly similar paralogs (FAM72A-D) unique to humans[1][2][4]. FAM72D contains a MIS18 (Yippee) domain homologous to domains in DNA maintenance proteins, and interacts with proteins involved in DNA replication and repair—including RPA complex proteins and uracil-DNA glycosylase (UNG)[1]. It is implicated in regulating DNA repair processes such as base excision repair and response to genotoxic stress, likely by forming homodimers or heterodimers with other FAM72 proteins and affecting chromatin recruitment during replication and DNA damage[1]. FAM72D is upregulated in multiple malignancies, including multiple myeloma and clear cell renal carcinoma, with its expression associated with cell cycle progression, tumor growth, and poor clinical prognosis[2][4].
