Overview

Family with sequence similarity 76 member A (FAM76A) is a nuclear-localized protein characterized by a coiled-coil domain and a Band7/PHB/SPFH domain of unknown function. It is broadly conserved among vertebrates and some invertebrates but is absent from fungi, plants, and prokaryotes. FAM76A is predicted to play a role in the regulation of transcription or RNA processing, supported by its nuclear location and association with nuclear speckle targeting motifs. Clinically, increased expression of FAM76A is associated with poor outcomes in ovarian cancer, possibly through interaction with the WTAP–HBS1L/FAM76A axis, and has been identified as a possible prognostic biomarker and therapeutic target in this context. Its precise molecular function and interactions are not fully characterized, and there are currently no drugs targeting FAM76A. Copy number variations disrupting the FAM76A locus have been linked to developmental anomalies, but no therapy-related safety concerns are established.
