Overview

Family with sequence similarity 83 member B (FAM83B) is a human protein, originally defined as a proto-oncogene and recognized for its critical role as an adaptor in the epidermal growth factor receptor (EGFR) signaling pathway, activating EGFR itself and its downstream effectors, including the RAS/MAPK, mTOR, and PI3K/AKT pathways[1][2][3][5]. FAM83B contains a distinctive N-terminal domain of unknown function (DUF1669), which mediates interactions with signaling effectors such as CRAF and casein kinase 1, facilitating oncogenic transformation, cell proliferation, and cell migration[1][2][4]. Overexpression of FAM83B has been found in various human cancers, where it correlates with increased tumor grade, dedifferentiation, and poor survival, indicating its role as a potential therapeutic target[1][2][3]. Although not itself a classical receptor, enzyme, or transcription factor, FAM83B functions centrally in signal transduction required for oncogenic activity and is actively studied for drug development efforts aimed at blocking multiple downstream proliferative signals[2][3][5]. There are currently no known drugs specifically targeting FAM83B, but its modulation could alter cellular sensitivity to existing targeted therapies, especially EGFR inhibitors[3]. Safety concerns include the risk of affecting normal cellular processes due to FAM83B’s involvement in fundamental signaling cascades.
