Overview

Family with sequence similarity 83 member C (FAM83C) is one of eight members of the FAM83 family, distinguished by the conserved DUF1669 domain—a motif with similarity to phospholipase D but lacking canonical enzymatic activity. FAM83C functions as a scaffolding protein regulating key kinases, including casein kinase 1 (CK1) isoforms, and promotes signal transduction by anchoring kinases to specific subcellular compartments. Its overexpression has been implicated in cancer cell transformation by facilitating aberrant activation of the MAPK signaling pathway, primarily through interactions with CRAF and possibly other components of EGFR/PI3K signaling axes. FAM83C is a novel oncogenic family member whose elevated expression is associated with various solid tumors and whose protein product is being considered as a candidate for future targeted therapeutics in oncology.
