Family with sequence similarity 83 member E (FAM83E) is one of eight members of the FAM83 protein family in humans, all of which are characterized by a highly conserved N-terminal domain of unknown function (DUF1669)[1][2][3]. FAM83E, like other FAM83 proteins, functions as a scaffold or anchoring protein and is capable of interacting with certain isoforms of casein kinase 1 (CK1), particularly CK1α, α-like, δ, and ε, via its DUF1669 domain. It is implicated in the spatial regulation of CK1 isoforms within cells and thereby participates in subcellular localization of signaling proteins[1]. Overexpression of FAM83E can promote oncogenic transformation of mammary epithelial cells, correlating with increased membrane localization of CRAF and activation of the MAPK signaling pathway, which is a pathway frequently dysregulated in cancers[2]. Elevated expression of FAM83E and other FAM83 members has been detected in a range of human cancers, where they may function as oncogenes and serve as candidate biomarkers for disease[2][1]. However, no drugs directly targeting FAM83E are currently known, and its utility as a clinical target or biomarker remains to be validated[1][2].
