Family with sequence similarity 83 member F (FAM83F) is a vertebrate-specific protein of the FAM83 family, defined by the conserved N-terminal DUF1669 domain, which possesses a pseudo-phospholipase D (PLD)-like motif but does not have PLD catalytic activity[2][4]. FAM83F localizes primarily to the plasma membrane and interacts selectively with casein kinase 1 alpha (CK1α), mediating canonical Wnt signalling and regulating CK1α subcellular localization[1][2]. Overexpression of FAM83F activates Wnt signalling, promoting cell proliferation, cell cycle progression, and inhibiting apoptosis in cancer cells, while knockout or loss attenuates these pathways and increases sensitivity to DNA damage, correlating with impaired autophagy and cellular degradation mechanisms[5][6]. High levels of FAM83F are linked to tumor progression and poor clinical outcomes in several cancers, especially lung adenocarcinoma and cervical cancer, making it a candidate therapeutic target and biomarker in oncology[4][5][6].
