Overview

Family with sequence similarity 89 member B (FAM89B) is a cytoplasmic adapter protein predicted to mediate transcription corepressor binding and modulate TGF-beta signaling by working in concert with SKI, preventing SMAD2 nuclear export in response to TGF-beta. It is hypothesized to regulate cell migration by targeting LIMK1 to the lamellipodia via CDC42BPA/B interaction, controlling actin dynamics critical for cellular protrusions. FAM89B is not a therapeutic target but participates in cellular processes relevant to cell polarity and movement. Disease associations are limited, with one mention of Cepacia syndrome but no evidence for direct targeting by drugs or utility as a clinical biomarker. It is predicted to be enriched in the cytoplasm and lamellipodium, involved with cell projections. FAM89B serves as an adapter for LIMK1 and CDC42BPA/B, impacting actin cytoskeleton remodeling. It is paralogous to FAM89A and not classically categorized among receptors, enzymes, or transcription factors. There is no evidence in current databases or literature for direct druggability, clinical targeting, or use as a diagnostic/prognostic biomarker.
