Overview

Fanconi anemia-associated protein 20 (FAAP20) is a 20 kDa protein that functions as an essential component of the Fanconi anemia (FA) core complex, a multi-protein E3 ubiquitin ligase vital for the repair of DNA interstrand crosslinks and maintenance of genome stability[1][2][3]. FAAP20 binds FANCA directly via a conserved C-terminal interaction, stabilizing FANCA and facilitating functional assembly and activity of the FA core complex. FAAP20 features a ubiquitin-binding zinc-finger domain (UBZ) that recognizes K63-linked polyubiquitin chains, required for DNA-damage-induced chromatin loading of the FA complex. Loss of FAAP20 impairs FANCI–FANCD2 monoubiquitination, deregulates DNA repair, and leads to genomic instability phenotypes characteristic of Fanconi anemia cells, including sensitivity to cross-linking agents and chromosomal aberrations[1][2][3][6]. FAAP20 also participates in homologous recombination and other homology-directed repair pathways. While not a direct therapeutic target itself, altered expression of FAAP20 has prognostic implications in certain cancers[6].
