Overview

Far upstream element-binding protein 1 (FUBP1) is a multifunctional, predominantly nuclear protein that binds single-stranded DNA (via FUSE upstream of the c-Myc gene) and also acts as an RNA-binding and splicing regulator[1][2][6][7]. FUBP1 plays a critical role in controlling c-Myc transcription, thus influencing cell growth, proliferation, and apoptosis[1][2][3][4]. It also regulates RNA stability and translation for various cellular and viral mRNAs, affecting diverse processes including development (neural, lung) and response to stress[1]. Aberrant FUBP1 expression or mutation is implicated in multiple cancers—either as an oncogene (when overexpressed) or a tumor suppressor (when lost, e.g., via 1p/19q codeletion in glioma)[2][3][4]. FUBP1 has also been linked to viral infection, as it binds viral RNAs and may facilitate viral replication[1][4]. Overall, it is recognized as a cancer-relevant transcription factor and nucleic acid-binding protein with context-dependent roles in oncogenesis and cell fate.
