Overview

Fas (CD95, APO-1) receptor is a transmembrane death receptor belonging to the TNF receptor superfamily, expressed on granulosa cells in ovarian follicles. Engagement of Fas by its ligand, FasL, leads to the induction of apoptosis via the caspase-8 pathway, playing a crucial role in the regulation of follicular atresia in the ovary. The signaling pathway is modulated by cytokines (e.g., interferon-γ), and cross-talks with other cellular pathways (e.g., ceramide-mediated signaling). Dysregulation of the Fas signaling pathway is implicated in several diseases, including cancer and reproductive disorders, and components of this pathway are being investigated as therapeutic targets and biomarkers.

Mechanism of action

Agonists induce apoptosis by triggering Fas-mediated cell death via caspase activation. Drugs may stabilize or enhance Fas/FasL interaction, or modulate downstream signaling (e.g., ceramide production)
