Fatty acid synthase I (FAS I) is a large, multifunctional enzyme complex responsible for the de novo synthesis of saturated fatty acids from simple precursors. It plays a central role in lipid metabolism and is essential for cell membrane formation, energy storage, and various signaling processes. In Mycobacterium tuberculosis, it is targeted by pyrazinamide, an anti-tuberculosis drug, via conversion to pyrazinoic acid, which inhibits mycobacterial FAS I, disrupting cell membrane synthesis and energy production pathways critical for bacterial survival under stress conditions. In mammals, altered expression or regulation is implicated in metabolic diseases like obesity, diabetes, and cancer.
