Overview

Fatty acid synthesis-related long non-coding RNA (FASRL), also called NQO1 divergent transcript (NQO1-DT), is a lncRNA reportedly located near the NQO1 locus but does not encode the canonical NQO1 protein. FASRL has been implicated in the regulation of cellular fatty acid synthesis by modulating metabolic enzyme activity and acts as an oncogenic factor in certain malignancies, notably hepatocellular carcinoma. Its expression is correlated with poor prognosis in cancer patients and may serve as a potential target for RNA-based therapeutics. The use of "NQO1-DT" may cause confusion with the NAD(P)H:quinone oxidoreductase 1 enzyme, a well-established drug and disease target in cancer and neurological disorders; however, they are distinct in function and molecular class[1][2].

Mechanism of action

Inhibiting the transcript (e.g. antisense, siRNA) to downregulate fatty acid synthesis and tumorigenesis

03

Biological functions

Regulation of fatty acid synthesisModulation of gene expressionInfluence on cellular proliferation and tumor growth
04

Disease associations

CancerMetabolic disease (potential)
05

Safety considerations

Limited evidence on safety due to lack of clinical targeting; risk relates primarily to unintended modulation of metabolic pathways and associated toxicities
06

Biomarkers

Overexpression of FASRL/NQO1-DT in tumor tissue (proposed as prognostic indicator)
