Overview

Faun269g is a novel ligand-gated ion channel identified by Fauna Bio through its Convergence platform, which leverages the unique physiological adaptations of hibernating mammals like the 13-lined ground squirrel (Ictidomys tridecemlineatus) (The Scientist, 2026; DeForest et al., 2025). In these animals, the target plays a critical role in providing multi-organ stress tolerance and protection against hypoxia and reperfusion injury during the extreme metabolic shifts of hibernation (Fauna Bio, 2025). In humans, genetic evidence from the UK Biobank and Million Veteran Program links variants in the Faun269g gene to heart failure with preserved ejection fraction (HFpEF) and pulmonary hypertension (PH) (DeForest et al., 2025). The target is modulated by FB-1083, a potent and selective small molecule antagonist that has demonstrated efficacy in preclinical models by improving diastolic function, reducing right ventricular systolic pressure, and protecting cardiomyocytes from mitochondrial stress (DeForest et al., 2025; Fauna Bio, 2025). By inhibiting Faun269g, FB-1083 aims to enhance cardiac output and mitochondrial efficiency, offering a potential first-in-class therapy for complex cardiovascular conditions (DeForest et al., 2025).
