Overview

Fc epsilon receptor I beta subunit (FcεRIβ) is a critical component of the high-affinity IgE receptor complex, primarily expressed on mast cells and basophils as part of a tetramer (FcεRIαβγ₂). The alpha chain binds IgE, while the beta subunit amplifies downstream signal transduction initiated by antigen-IgE crosslinking, leading to cell activation and release of allergic and inflammatory mediators. FcεRI also plays a role in immune defense against parasitic infections and can be modulated by drugs such as omalizumab. Genetic variation and cell-specific expression of FcεRIβ are increasingly studied as biomarkers for allergy severity and therapeutic response.

Mechanism of action

Omalizumab: Binds circulating IgE, preventing it from engaging FcεRI, reducing receptor activation and allergic inflammation. DARPins and fusion proteins: Interfere with FcεRI signaling or co-engage inhibitory FcγRIIb, blocking mast cell/basophil activation.
