Fc gamma receptor IV (FcγRIV)

Target
FcγRIV
Molecular classification
Receptor, Fc receptor, Immunoglobulin superfamily
01

Overview

Fc gamma receptor IV is a murine immune cell receptor belonging to the immunoglobulin superfamily, structurally characterized by extracellular Ig-like domains and an intracellular ITAM signaling motif via association with the Fc receptor gamma chain. It binds the Fc portion of IgG2a and IgG2b with intermediate affinity and mediates key effector functions such as phagocytosis, antigen presentation, and inflammation. FcγRIV plays a central role in antibody-dependent immune responses and contributes to both protection and immunopathology in mouse models. This receptor does not have a direct human ortholog; in humans, related functions are performed by FcγRIIIA (CD16a)[5][7].

Mechanism of action

Drugs or antibodies that engage FcγRIV induce ITAM-mediated signaling, triggering phagocytosis, cytokine release, and cell-mediated cytotoxicity

05

Safety considerations

Manipulating FcγRIV signaling can provoke excessive inflammation or tissue damage due to its strong activation potential; animal studies highlight risk of immune hyperactivation
06

Interacting drugs

Experimentally, monoclonal antibodies and immune complexes that bind IgG2a and IgG2b subclasses interact with FcγRIV
