The Fc receptor gamma chain (FcRγ) is a critical signaling subunit associated with several classes of immunoglobulin Fc receptors, particularly those for IgG (FcγRs) and IgE (FcεRI). It acts as an essential adaptor protein that enables signal transduction and surface expression for various activating Fc receptors. Upon engagement of the associated receptor by immune complexes or antibody-coated targets, the ITAMs on the gamma chain become phosphorylated, initiating intracellular signaling cascades. The gamma chain is required for proper assembly and cell surface expression of certain activating Fc receptors. It plays central roles in both innate and adaptive immunity, including enhancing clearance of pathogens through phagocytosis, mediating ADCC, and regulating inflammation.
