Overview

FCH and double SH3 domains protein 1 (FCHSD1) is a cytosolic protein containing an amino-terminal F-BAR domain and two C-terminal SH3 domains, distantly related to the formin-binding proteins FNBP1/FNBP2[3][1]. FCHSD1 functions as an actin-regulatory factor, primarily expressed in cuticular plates of sensory hair cells where it influences actin filament organization and maintenance through its interaction with Sorting Nexin 9 (SNX9), enhancing SNX9-mediated, Arp2/3-dependent actin polymerization[1]. FCHSD1 also forms complexes with the transcription factor NRF2 and SNX9 in non-neuronal cells, suppressing NRF2 nuclear translocation, with functional implications in regulating oxidative stress response and emphysema development: its genetic loss in mice protects against lung inflammation and apoptosis in experimental COPD[2]. There are no known drugs targeting FCHSD1, and its direct role as a therapeutic target or as a biomarker remains unestablished.
