Overview

The envelope glycoprotein (Env) of the feline immunodeficiency virus (FIV) is a critical viral protein responsible for mediating the entry of the virus into host cells. Like other lentiviruses, FIV encodes a single Env precursor that is cleaved into two non-covalently associated subunits: the surface glycoprotein (SU, also known as gp95) and the transmembrane glycoprotein (TM, also known as gp35). The SU binds first to CD134 on target cells—a primary receptor unique to FIV among lentiviruses, then engages CXCR4 as a coreceptor. After receptor engagement, conformational changes in TM mediate fusion between viral and cellular membranes—enabling viral entry. Variable regions within SU—especially V3 and V5 loops—are key determinants for coreceptor usage and immune evasion.
