Overview

FERM and PDZ domain-containing protein 1 (FRMPD1) is a large, multi-domain scaffold/adaptor protein containing a FERM domain and a PDZ domain.[2][4] It functions as a versatile adaptor protein that modulates key signaling pathways and cellular processes, notably by participating in the regulation of cell polarity, spindle orientation (via interaction with LGN and AGS3), and Hippo pathway activation through binding to WWC3.[2][4] In cancer biology, especially in non-small cell lung cancer, FRMPD1 acts as a tumor suppressor by activating the Hippo pathway, inhibiting YAP nuclear translocation, thereby reducing cell proliferation and invasion; its reduced expression correlates with poor prognosis.[4] FRMPD1 is expressed in a tissue-specific manner, including documented retinal-specific expression, but is not known to be a direct therapeutic target or receptor for any approved drugs.[2][4][6]
