Overview

FERM and PDZ domain-containing protein 2 (FRMPD2) is a multi-domain peripheral membrane protein found predominantly in epithelial cells[1][2][3][4]. Its structure comprises an N-terminal KIND (Kinase non-catalytic C-lobe domain), a FERM (Four-point-one, ezrin, radixin, moesin) domain responsible for membrane localization, and three PDZ domains essential for targeting to junctional regions and scaffolding functions[1][3]. The protein is recruited to cell-cell junctions in an E-cadherin-dependent manner and is selectively localized at the basolateral membrane in polarized epithelial cells[1][2]. It interacts with catenin family members (such as δ-catenin, p0071, and ARVCF) through its PDZ domains, supporting tight junction formation and possibly influencing cell polarity[1]. FRMPD2 may also play a role in synapse formation and immune signaling, but these roles are less clearly established[3][4]. Disease associations are limited, mainly involving rare genetic syndromes or conditions linked to cell junction integrity and polarity[3]. No drugs or biomarkers targeting FRMPD2 are known, and it is not classified as a therapeutic target.
