Overview

FERM domain-containing protein 3 (FRMD3) is a member of the FERM domain protein superfamily, characterized by a conserved ~150 amino acid FERM domain found in cytoskeletal adaptor proteins that link the cell membrane to the cytoskeleton[3][5]. FRMD3 acts as a tumor suppressor, notably in breast cancer, where it inhibits cell proliferation, migration, invasion, and metastasis, partly by promoting polyubiquitin-mediated degradation of vimentin and by regulating the Notch signaling pathway through destabilization of Disheveled-2 and modulation of epithelial cell fate[2][4]. FRMD3's low expression is associated with increased tumorigenic potential and poorer prognosis in breast cancer patients[4]. Its primary biological functions involve the maintenance of cytoskeletal architecture, control of cell morphology, and regulation of membrane-protein interactions[2][3][5].
