Overview

FRMD6 (FERM domain-containing protein 6), also known as Willin and expanded homolog, is a 622 amino-acid cytoskeletal protein characterized by a FERM domain at the N-terminus and is a key regulator of cell polarity, adhesion, and the Hippo signaling pathway[1][2][4][5]. Its FERM domain mediates interactions with transmembrane proteins and the plasma membrane, allowing it to influence cell junctions and membrane domain size[2][3]. FRMD6 activates Hippo pathway kinases MST and LATS, thereby antagonizing the oncogenic YAP transcriptional regulator and promoting cellular senescence and tumor suppression[1]. FRMD6 expression is upregulated during cellular senescence and in aging tissues, and its gene has been implicated in neurodegeneration and cognitive impairment in clinical studies[2]. It is mainly localized at cell junctions, cytoplasm, and plasma membrane and displays conserved expression across vertebrates[2][4][5].
