FERM domain-containing protein 7 (FRMD7) is a cytoskeletal regulatory protein most notable for its role in the development of nerve cells in regions of the brain that control eye movement, as well as in the retina[1][3][5]. It belongs to the FERM domain protein family, which links the plasma membrane to the actin cytoskeleton and participates in signal transduction[1][4]. FRMD7 interacts with the Rho GDP dissociation inhibitor alpha (RhoGDIα) and can activate Rac1 signaling, thereby influencing actin organization and neurite extension during neuronal development[1][3]. Mutations in FRMD7, especially those disrupting either its FERM or C-terminal domains, impair its function and are associated with X-linked idiopathic congenital nystagmus, a disorder characterized by involuntary, repetitive eye movements[1][5]. The gene encoding FRMD7 is most abundantly expressed in the midbrain and cerebellum, and its dysfunction is associated with abnormal development of nerve cells critical for ocular motor control[5]. While FRMD7 is a key protein in neurodevelopment, it is not currently considered a therapeutic drug target, and there are no known interacting drugs or widely used clinical biomarkers beyond its genetic mutations in disease diagnosis[5].
