Fetal and adult testis expressed 1 (FATE1) is a protein-coding gene encoding a cancer/testis antigen predominantly expressed in testis (fetus and adult) and aberrantly upregulated in multiple human cancers, including Ewing sarcoma and hepatocellular carcinoma[3][4][6][7]. FATE1 is a mitochondrial membrane protein involved in the regulation of endoplasmic reticulum-mitochondria coupling and apoptotic signaling[3][7]. It supports tumor cell fitness by controlling the protein level of pro-apoptotic factors such as BIK through an interaction with the E3 ligase RNF183[2][3]. In Ewing sarcoma, FATE1 is a direct target of the EWSR1-FLI1 fusion oncogene, and its function is required for cancer cell survival and anchorage-independent growth[1][2]. Given its restricted normal expression and immunogenicity, FATE1 is being explored as a potential immunotherapy target, though no therapeutic agents or drugs directly targeting FATE1 are currently approved or in clinical use[2][3].
