Overview

Fetuin-B is a secreted glycoprotein, part of the fetuin family within the cystatin superfamily, and acts as a highly selective and potent inhibitor of astacin-family metalloendopeptidases, particularly ovastacin. By preventing premature zona pellucida hardening in mammalian oocytes, it is critical for female fertility. Beyond reproductive roles, fetuin-B may participate in diverse biological processes including bone metabolism, insulin signaling regulation, and modulation of inflammatory responses. It is distinct from, but structurally related to, fetuin-A. Human FETUB is encoded on chromosome 3, with protein encoded in multiple vertebrates. No approved drugs target fetuin-B directly, and its main significance lies in endogenous regulation rather than as a traditional therapeutic target.
