Overview

Fez family zinc finger protein 1 (FEZF1) is a transcription factor containing C2H2-type zinc fingers, encoded by the FEZF1 gene. It is highly conserved across vertebrates and is critical for proper neural development, including forebrain patterning, differentiation of neural stem cells, olfactory sensory neuron maturation, and migration of gonadotropin-releasing hormone (GnRH) neurons. FEZF1 acts as a transcriptional repressor and activator, modulating genes such as Hes5 to promote neurogenesis and activating Wnt signaling pathway targets to enhance cell proliferation, growth, and migration in cancer cells. Aberrant FEZF1 expression has been implicated in several cancers—including cervical, gastric, and colorectal cancers—where it may promote malignant progression. Mutations or dysregulation of FEZF1 are involved in neurodevelopmental syndromes such as Kallmann syndrome, with loss of function resulting in impaired forebrain segmentation and GnRH neuron migration deficits.
