Overview

FGD5 antisense RNA 1 (FGD5-AS1) is a long non-coding RNA (lncRNA) located on chromosome 3, transcribed antisense to the FGD5 gene, but not coding for protein[7]. It is implicated in several human diseases. In cancer, FGD5-AS1 generally acts as an oncogene, promoting proliferation, migration, invasion, and stemness of tumor cells—including in non-small cell lung cancer and colorectal cancer—via functioning as a competing endogenous RNA (ceRNA) that sequesters microRNAs, leading to upregulation of oncogenic proteins[5][6][7]. In cardiovascular disease, FGD5-AS1 has been shown to protect cardiomyocytes against hypoxia-induced oxidative damage and apoptosis, partly by regulating the RORA gene via miR-195-5p, suggesting a protective role in acute myocardial infarction[1][2][3]. FGD5-AS1 is also reported to participate in inflammatory responses and may play roles in other disease contexts. At present, it is not classified as a conventional therapeutic target such as a receptor, enzyme, transporter, or transcription factor, and no drugs are known to interact directly with FGD5-AS1, although its roles as a ceRNA and gene expression regulator make it of substantial research interest in biomarker and mechanistic studies[7].
