Overview

FGF13 antisense RNA 1 (FGF13-AS1) is a long non-coding RNA (lncRNA) transcribed antisense to the FGF13 gene. Unlike protein-coding genes, FGF13-AS1 does not encode a protein but instead functions primarily through RNA-based mechanisms. In cancer biology, particularly in breast cancer, FGF13-AS1 acts as a tumor suppressor: its expression is decreased in tumor tissue compared to normal tissue, and low levels are associated with poor prognosis. Mechanistically, FGF13-AS1 inhibits cancer cell proliferation, migration, and invasion by impairing glycolysis and restricting stemness properties. It achieves this by binding insulin-like growth factor 2 mRNA-binding proteins (IGF2BPs), disrupting their interaction with MYC mRNA, and thereby reducing MYC mRNA stability and protein levels. MYC participates in a feedback loop by transcriptionally repressing FGF13-AS1, indicating a tightly regulated axis. More broadly, FGF13-AS1 and similar antisense lncRNAs modulate gene expression by influencing mRNA stability and translation dynamics, and may act as epigenetic or post-transcriptional regulators in various physiological and pathological processes. There is currently no evidence that FGF13-AS1 is a direct therapeutic target for drugs, but its modulation may present new opportunities as a biomarker or adjunct target, primarily in oncology.
