Overview

FHF complex subunit HOOK-interacting protein 1B (FHIP1B) is a protein component of the FTS/Hook/FHIP (FHF) complex, which promotes vesicle trafficking and/or vesicle fusion through interactions with the HOPS (homotypic fusion and protein sorting) complex[1][6]. FHIP1B functions as a cytoplasmic cargo adaptor, specifically linking the dynein motor complex to early endosomes via direct binding to Rab5B[2]. In cells, FHIP1B associates with other adaptor proteins Hook1 and Hook3 to facilitate targeting and motility of early endosomes along microtubules, contributing to endosomal organization and trafficking[2]. There is no current evidence or consensus that FHIP1B is a direct therapeutic target, nor are there any drugs known to interact with it. Disease associations are rare and largely genetic, without direct evidence for therapeutic modulation[1][2].
