Overview

The interaction between the fibrin E1 fragment and vascular endothelial (VE)-cadherin is a key molecular event implicated in angiogenesis, inflammation, and vascular integrity. VE-cadherin is an endothelium-specific adhesion protein located at intercellular junctions of endothelial cells. The binding site for the fibrin E1 fragment on VE-cadherin has been mapped to its extracellular domains EC3 & EC4. On the fibrin side, the E1 fragment corresponds to a region within fibrin(ogen) that includes both βN-domains. Specific residues within these βN-domains—particularly His16 and Arg17—are essential for binding to VE-cadherin. This interaction triggers capillary-like structure formation by endothelial cells and modulates inflammatory responses by affecting endothelial permeability.
