Overview

The Fibrin P2 epitope is a cryptic, pro-inflammatory sequence exposed on the fibrinogen γ chain (amino acids 377–395) after conversion of fibrinogen to insoluble fibrin by thrombin cleavage. This epitope is not accessible in soluble fibrinogen and becomes available only upon formation of cross-linked, insoluble fibrin. Upon exposure, the P2 epitope binds to CD11b/CD18 (complement receptor 3/CR3/Mac-1/αMβ2 integrin) and CD11c/CD18 (CR4/αXβ2 integrins) receptors found on microglia, macrophages, neutrophils, and dendritic cells, triggering innate immune activation and potentially contributing to neurodegenerative diseases.
