Overview

Fibroblast growth factor 17 (FGF17) is a secreted member of the FGF family, playing a critical role as a signaling molecule in embryonic brain development, vascular formation, bone development, and tissue repair. FGF17 is expressed in the cerebellum, cerebral cortex, and midline embryonic brain structures and is essential for normal development of the central nervous system and vasculature. Mutations in the FGF17 gene are associated with some forms of congenital hypogonadotropic hypogonadism. FGF17 signals through high-affinity fibroblast growth factor receptors (FGFRs), predominantly activating pathways involved in cell proliferation and differentiation. No clinically approved drugs directly target FGF17, but aberrant signaling has been linked to cancer progression.

Mechanism of action

Ligand for fibroblast growth factor receptors (FGFRs), especially type 2. Activates downstream signaling via tyrosine kinase FGFRs, inducing RAS/MAPK, PI3K/AKT, and PLCγ pathways.
