Fibroblast growth factor binding protein 1 (FGFBP1) is a secreted, extracellular carrier protein that binds fibroblast growth factors (FGFs), particularly FGF-1 and FGF-2, facilitating their release from the extracellular matrix and potentiating their mitogenic, angiogenic, and cell migration-inducing activities on target cells. FGFBP1 plays a crucial role in tissue repair, wound healing, and the pathophysiology of diverse cancers by acting as an "angiogenic switch" and enhancing tumor progression through FGF-driven signaling. FGFBP1 also modulates immune-related pathways and interacts with immune checkpoint proteins like PD-L1, influencing tumor microenvironment and immune escape. In cardiovascular biology, FGFBP1 sensitizes blood vessels to angiotensin II, impacting vascular tone and elevating blood pressure through FGF-dependent MAPK signaling. Its expression is upregulated in various pathological states, including multiple cancer types, inflammatory lesions, and hypertension, and it is being evaluated as a biomarker for prognosis and therapeutic responsiveness.
