Overview

Fibroblast growth factor receptor 1 oncogene partner 2 (FGFR1OP2) is a cytoskeleton-associated coiled-coil domain protein that plays a physiological role in wound healing by regulating fibroblast movement, contractility, and efficient tissue repair with minimal scarring. FGFR1OP2 dimerizes and localizes to the cytosol and cytoskeleton. Under normal conditions, its main functions are in wound healing and possibly response to thyroid hormone signaling during development. However, when involved in rare chromosomal translocations leading to the formation of FGFR1OP2-FGFR1 fusion proteins, it acts as an oncogenic partner, promoting constitutive tyrosine kinase activity and driving aberrant cell proliferation—resulting in aggressive hematological malignancies such as myeloproliferative neoplasms and lymphoblastic lymphoma. FGFR1OP2 does not serve as a direct drug target or receptor, but its fusion with FGFR1 may create oncogenic drivers that could theoretically be targeted by FGFR kinase inhibitors in the disease context. Its standalone protein function remains under investigation, and it is not established as a classic therapeutic target.
