Overview

The fibroblast growth factor receptor (FGFR) family is a group of receptor tyrosine kinases that play essential roles in cell signaling, development, tissue homeostasis, and disease. The family consists of four main members: FGFR1, FGFR2, FGFR3, and FGFR4. A fifth member (FGFR5) has been identified but lacks the intracellular kinase domain present in the others. FGFRs bind to fibroblast growth factors (FGFs), and upon ligand binding, dimerize and transautophosphorylate, initiating downstream signaling cascades involved in proliferation, differentiation, migration, survival, angiogenesis, and metabolic regulation. Dysregulation or mutation of FGFR genes is implicated in various diseases, including cancers and congenital disorders.
