Fibronectin 1 (FN1) is a high-molecular-weight glycoprotein found in the extracellular matrix and plasma that plays a pivotal role in cell adhesion, migration, and differentiation (UniProt P02751). It serves as a primary ligand for several integrin receptors, particularly integrin alpha-5 beta-1, which mediates cell adhesion and signaling (NCBI Gene 2335). In the immune system, fibronectin is crucial for the migration of leukocytes and the phagocytic activity of macrophages (PubMed PMID: 15549731). The phrase 'fibronectin-mediated adhesion and internalization' often refers to the mechanism by which pathogens or viral vectors enter host cells (PubMed PMID: 11544357). Recombinant fibronectin fragments, such as RetroNectin, are extensively utilized in clinical settings to enhance the transduction efficiency of retroviral and lentiviral vectors into T cells for immunotherapy (Takara Bio). Fibronectin is also involved in wound healing and tissue repair, where it acts as a scaffold for cell migration (PubMed PMID: 23681448). In disease states, elevated fibronectin levels are associated with cancer metastasis and the development of organ fibrosis (PubMed PMID: 25108357). Therapeutic agents targeting the fibronectin-integrin axis, such as volociximab, have been investigated for their anti-angiogenic and anti-tumor properties (ClinicalTrials.gov). Monitoring fibronectin levels, such as oncofetal fibronectin, serves as a biomarker in obstetric and oncological contexts (PubMed PMID: 17210598).
