Overview

Fibronectin type III domain-containing protein 3B (FNDC3B) is a membrane-associated protein characterized by nine fibronectin type III (FNIII) domains and a single transmembrane domain near the C-terminus[1][2][3]. It is primarily localized to cellular membranes, especially the endoplasmic reticulum and Golgi apparatus, with correct localization essential for its biological and oncogenic functions[1][2]. FNDC3B is involved in cell adhesion, migration, proliferation, and signaling, and has a prominent role in promoting epithelial–mesenchymal transition (EMT), activating oncogenic pathways (such as PI3K/Akt, Rb, and TGF-β), and supporting tumor maintenance and metastasis, especially in hepatocellular carcinoma and other cancers[2][3][5]. Its overexpression is correlated with poor prognosis in several cancer types. FNDC3B also regulates adipocyte and osteoblast differentiation, and is implicated in ER stress response and protein processing pathways[1][3][5].
