Overview

Fibrous sheath-interacting protein 1 (FSIP1) is a structural protein that forms part of the fibrous sheath in sperm flagella, essential for sperm motility and integrity. In cancer cells, FSIP1 is aberrantly overexpressed where it can stabilize microtubule networks, interact with cancer/testis antigens, and promote cell proliferation and metastatic features. Elevated FSIP1 levels are strongly correlated with poor outcomes in breast and non-small cell lung cancer patients, and FSIP1 mediates chemoresistance by modulating the stability of multidrug resistance protein 1 (MRP1). FSIP1 is under investigation as both a pathological biomarker and as a therapeutic target, particularly for combating drug resistance and progression in certain cancers.

Mechanism of action

Drugs targeting FSIP1 may sensitize tumor cells to chemotherapeutic agents by reducing FSIP1 expression or function, thereby increasing cellular drug accumulation and decreasing resistance (e.g., docetaxel sensitivity improves upon FSIP1 knockout)
