Overview
Filaggrin is a key structural protein essential for the formation and maintenance of the skin barrier. It is primarily found in the outermost layers of the epidermis, where it plays a critical role in aggregating keratin intermediate filaments, facilitating cell flattening, and contributing to skin hydration. Loss-of-function mutations in FLG are strongly associated with ichthyosis vulgaris and atopic dermatitis/eczema. After fulfilling its structural role, filaggrin is degraded into free amino acids components collectively known as "natural moisturizing factor" (NMF).
