Overview

Filamin-binding LIM protein 1 (FBLIM1) is a cytoskeletal adaptor protein encoded by the FBLIM1 gene in humans. It features an N-terminal filamin-binding domain, a central proline-rich region, and multiple C-terminal LIM domains. FBLIM1 localizes chiefly at cell junctions, where it connects cell adhesion complexes to the actin cytoskeleton, helping to regulate cell shape, adhesion, and movement. It also localizes in the nucleus and may influence differentiation processes through interactions with transcription factors. FBLIM1 is implicated in the assembly and stabilization of actin filaments, promotes integrin activation, and helps organize signaling complexes that regulate cytoskeletal dynamics and cell-cell contacts. Variants in FBLIM1 are linked to certain diseases such as Kindler syndrome and osteomyelitis.
