Overview

Filamin-C (FLNC) is a large, dimeric actin-binding protein predominantly expressed in striated muscle, including cardiac and skeletal muscle. It is composed of an N-terminal actin-binding domain followed by 24 immunoglobulin-like repeats and is essential for maintenance and structural integrity of the Z-disc in the sarcomere. FLNC cross-links actin filaments, anchors membrane proteins to the cytoskeleton, and acts as a mechanosensor to transmit force during muscle contraction. Unique features include specialized insertions and regulated dimerization for muscle-specific functions. Mutations in FLNC cause various muscle diseases, mainly cardiomyopathies and myofibrillar myopathies, highlighting its importance in muscle health and disease.
