Overview

Filovirus antigens encoded by MVA-BN-Filo consist of a multivalent set of proteins designed to provide broad protection against several deadly hemorrhagic fever viruses. Specifically, the MVA-BN-Filo vaccine vector expresses the glycoproteins (GP) of Ebola virus (Mayinga strain), Sudan virus (Gulu strain), and Marburg virus (Musoke strain), along with the nucleoprotein (NP) of the Tai Forest virus. These glycoproteins are the primary targets for neutralizing antibodies as they facilitate viral attachment and entry into host cells, while the nucleoprotein is included to enhance the breadth of the T-cell response. In a clinical setting, these antigens are delivered via a viral vector (Modified Vaccinia Ankara) as part of a heterologous prime-boost regimen, typically following a prime dose of Ad26.ZEBOV. This approach aims to generate durable humoral and cellular immunity to prevent Ebola and Marburg virus diseases.

Mechanism of action

The antigens are expressed by the MVA-BN vector following vaccination, where they are presented to the immune system to induce virus-specific neutralizing antibodies and T-cell mediated immune responses against multiple filovirus species.
