Overview

Fin bud initiation factor homolog (FIBIN) is a secreted protein with evolutionarily conserved roles in embryonic development and adult tissue function. In zebrafish, FIBIN is essential for the initiation of pectoral fin buds, acting upstream of key regulatory genes including tbx5 and functioning downstream of retinoic acid and Wnt signaling[1][4]. In mammals, FIBIN is expressed in a variety of tissues, prominently in the cerebellum and skeletal muscle, and is characterized by a cleavable signal peptide and glycosylation. Most FIBIN protein is retained in the endoplasmic reticulum unless secreted by an as-yet-unknown mechanism[1][2][5]. FIBIN has also been implicated in muscle homeostasis, where dysregulation (especially due to microRNA interference such as miR-141-3p) can impair mitochondrial function and myogenic differentiation. Evidence also links FIBIN expression with suppression of metastasis in non-small cell lung cancer (as a DNA methylation marker) and associations with rare muscle and metabolic diseases; however, FIBIN is not considered a mainstream therapeutic target such as a receptor, enzyme, or ion channel[1][5][9].
