Overview

Firre intergenic repeating RNA element (FIRRE) is a long non-coding RNA (lncRNA) located on the X chromosome (chrX:131702650-131830643, hg38). FIRRE is not translated into protein, as shown by multiple protein-coding potential analyses. Its transcript coats chromosomes and is retained in the nucleus via interaction with heterogeneous nuclear ribonucleoprotein U (hnRNPU). FIRRE serves as a scaffold for chromatin organization—it mediates chromatin looping and recruits histone modifiers, anchoring the inactive X chromosome to the nucleolus and maintaining H3K27me3 repressive methylation. FIRRE is implicated in trans-chromosomal interactions and has documented roles in regulation of gene expression relevant to pluripotency, adipogenesis, and innate immunity by modulating mRNA stability. There are currently no direct drug interactions or established biomarker or safety data for FIRRE.
