FKBP prolyl isomerase family member 6 (FKBP6) is a member of the FK506 binding protein (FKBP) family and functions as an immunophilin related to protein folding and trafficking[3]. Unlike some other FKBPs, FKBP6 is considered catalytically inactive as a peptidyl-prolyl cis-trans isomerase, though it shares the family’s structural domain[3]. FKBP6 has a critical role in spermatogenesis and maintenance of germline genome integrity through repression of transposable elements in germ cells, acting through the piRNA metabolic pathway and forming complexes with Piwi proteins for transposon silencing[3]. The gene is located in a region that can be deleted in Williams-Beuren syndrome, and its genetic defects are associated with specific forms of male infertility[3]. FKBP6 does not appear to be a direct therapeutic target, nor are drugs currently known to interact with it, but its loss-of-function or mutation is important in reproductive biology and genomic regulation.
