Flagellar hook protein FlgE is a key structural component of bacterial flagella, specifically forming the hook that connects the basal body to the filament. This flexible "universal joint" transmits torque from the motor apparatus in the basal body to rotate the filament, enabling bacterial motility. The flgE gene encodes this subunit, which is essential for proper flagellum assembly, swimming ability, and biofilm formation. In some species such as Helicobacter pylori, multiple forms of FlgE exist with distinct but complementary roles in organizing polar flagella. Beyond its mechanical function, FlgE can also act as an antigen and elicit pro-inflammatory responses during infection. While it is not a direct therapeutic target like enzymes or receptors commonly targeted by drugs, understanding its structure and function may inform antibacterial drug development aimed at impairing bacterial motility or colonization[1][4][5][6].
