Overview

The flavivirus envelope (E) protein is a structural glycoprotein found on the surface of flaviviruses, essential for viral entry and playing multiple roles throughout the viral life cycle. It mediates attachment to host receptors, membrane fusion during entry, virion assembly, and egress. The E protein consists of three domains (EDI, EDII, EDIII) connected by flexible hinges. EDII contains the fusion loop, and EDIII is involved in receptor binding. Glycosylation affects viral attachment, cell entry, virion stability, replication efficiency, and pathogenesis. The E protein is a major target for antiviral drug development and vaccine design due to its surface exposure and immunogenicity.
