The FCU1 enzyme is a bifunctional fusion protein comprised of cytosine deaminase and uracil phosphoribosyltransferase activities. It converts the prodrug flucytosine (5-FC) into the cytotoxic chemotherapeutic agent fluorouracil (5-FU) within target cells. This conversion is a key component of gene-directed enzyme prodrug therapy (GDEPT) strategies for cancer treatment, allowing for targeted tumor cell killing while minimizing systemic toxicity associated with direct fluorouracil administration.
Enzymatic conversion of flucytosine to fluorouracil, followed by phosphorylation into cytotoxic nucleotide analogs that inhibit DNA and RNA synthesis.
