Fludarabine phosphate is a water-soluble prodrug used in the treatment of hematologic malignancies. It is converted to its active form intracellularly, which inhibits DNA synthesis and induces apoptosis in cancer cells.
Fludarabine phosphate is a prodrug that is dephosphorylated to 2-fluoro-ara-A (F-ara-A), which is then phosphorylated to the active metabolite 2-fluoro-ara-ATP (F-ara-ATP). F-ara-ATP inhibits DNA polymerase alpha, ribonucleotide reductase, DNA primase, and DNA ligase, and is incorporated into DNA, causing chain termination.
